| Description: | DNA profiling studies, performed by ATCC, revealed that STR patterns of the endothelial line ECV-304 and the human bladder line T24 were very similar, suggesting that ECV-304 was a derivative of T24. Furthermore, ATCC karyotypes of the two lines show two shared-marker chromosomes. Thus, ECV-304 mopst probably is a derivative of T24, a line that was developed a couple of years earlier. It is important to emphasize that all stocks of ECV show similar properties, not just those from ATCC. It is clear that the contamination took place before the cell banks independently received initial stocks. Independent laboratories have reported that ECV showed some biomarkers expected of endothelial cell lines, namely Factor VIII, tubule formation on Matrigel, and/or Weibel-Palade bodies. Studies to determine if T24 exhibits any such markers are underway. |
| Culture Medium: | Medium 199 supplemented with 2 mM L-glutamine and 10% fetal bovine serum (MG-100, CLS order number 821100). |
| Subculturing: | Remove medium and rinse the adherent cells using PBS without calcium and magnesium (3-5 ml PBS for T25, 5-10ml for T75 cell culture flasks). Add TrypleExpress (1-2ml per T25, 2.5ml per T75 cell culture flask), the cell sheet must be covered completely. Incubate at 37°C for 10 minutes. Carefully resuspend the cells, the addition of medium is optional but not necessary, and dispense into new flasks which contain fresh medium. |
